It is believed that genetic and environmental risk factors cause schizophrenia by changing normal developmental pattern of the brain. The exact nature of neurodevelopmental abnormalities remains unknown; however, they are presumed to lay down faulty neural circuits impacting brain chemistry and functions in adulthood. Anatomical abnormalities in the temporal lobe including the hippocampal formation are widely reported in persons affected with schizophrenia. To understand the importance of normal development of hippocampus in adult behaviors, we introduce defined hippocampus lesions in rats at neonatal ages. The neonatal ventral hippocampus lesioned (NVHL) rats show a variety of schizophrenia related behaviors when they reach post-pubertal age. Our work as well as studies by other investigators has shown a fundamental abnormality in the structure and function of the prefrontal cortex in NVHL rats. In this seminar, I will discuss our results showing possible neurochemical and molecular mechanisms that may be responsible for delayed behavioral changes after NVH lesions. It is hoped that these investigations will help us better understand the neurobiological mechanisms underlying schizophrenia
